The PPTg is rich in cholinergic neurons (Mesulam et al., 1983) and several studies have identified serotonergic and glutamatergic terminals in the structure (Mazzei-Silva et al., 2014; Clements and Grant, 1990; Roseberry et al., 2016). PPTg glutamatergic neurons encode locomotion and speed by highly specific connection with basal ganglia (Roseberry et al., 2016).

Subpopulations of these cell classes expressed calcium-binding proteins (calbindin, calretinin, and parvalbumin) and they are heterogeneously distributed within the PPT (Martinez-Gonzalez et al., 2012).
