Glutamate-like immunoreactivity in neurons of the laterodorsal tegmental and pedunculopontine nuclei in the rat

Abstract

Studies of the pedunculopontine (PPT) and laterodorsal tegmental (LDT) nuclei in the mesopontine tegmentum have emphasized the organization and projections of the cholinergic neurons. We report here that exhibiting glutamate immunoreactivity are present in both the LDT and PPT. These glutamatergic neurons are interspersed among the cholinergic neurons within both nuclei with no apparent segregation. These data raise the possibility that excitatory amino acids contribute to the functions of the LDT and PPT.

    Functional PPTg inactivation decreases the antinociceptive phenomenon during generalized tonic-clonic seizures (Mazzei-Silva et al., 2014). The PPTg is rich in cholinergic neurons (Mesulam et al., 1983) and several studies have identified serotonergic and glutamatergic terminals in the structure (Mazzei-Silva et al., 2014; Clements and Grant, 1990; Roseberry et al., 2016). PPTg glutamatergic neurons encode locomotion and speed by highly specific connection with basal ganglia (Roseberry et al., 2016).

    Anatomy. The PPT and LDT are located within the caudal cholinergic column (Ch5 and Ch6) and contain a heterogeneous population of cholinergic, glutamatergic and GABAergic neurons (Clements and Grant, 1990; Wang and Morales, 2009; Ford et al., 1995). Subpopulations of these cell classes expressed calcium-binding proteins (calbindin, calretinin, and parvalbumin) and they are heterogeneously distributed within the PPT (Martinez-Gonzalez et al., 2012).
